Your idea is snowflakey and doesn't scale with new species added. Eh, why not, it would make it easier to identify the person who drawn it (can IPCs make runes out of oil?) here is the flipside, lets say there is a engineer cultist and a vox cultist from a different department, the engineer lets the vox in and the vox makes a rune for some reason and leaves, then security comes but they cant pin it on the engineer since its vox blood but not the engineers blood.
